SUMMARY:

The snail Biomphalaria glabrata is the intermediate host of the parasite Schistosoma mansoni; the only molluscicide available to control this disease has shown high cost and low selectivity, which leads to the search for new alternatives. The objective of this work was to evaluate the toxicity of the saline extract and lectin-enriched fraction of Parkia pendula seeds on embryonic and adult stages of B. glabrata, cercaria of S. mansoni and the ecotoxicity against Artemia salina. The saline extract (ES) obtained from the suspension in 0.15 M NaCl and the fraction (FR) with 0-60% ammonium sulfate were evaluated by thin layer chromatography, high performance liquid chromatography and physicochemical analyses. Blastula, gastrula, trochophora, veliger and Hippo stage embryos and adult snails were exposed (24 h) to ES and FR and reproductive parameters (fecundity and fertility) and survival (seven days after exposure), as well as cytotoxicity and genotoxicity were analyzed. In addition, the effects of fenced and environmental safety using A. salina were performed. ES showed only reducing sugars (0.86 mg/mL), while FR did not reveal any metabolite analyzed. The protein content of EP was 5.1 mg/mL, hemagglutinating activity (HA) of 64 and specific HA (SHA) of 12.54, while FR contained 0.91 mg/mL of protein, HA 128 and SHA 140, 65. ES and FR showed toxic effects for embryos (LC90 of 464.25; 479.62; 731.28; 643.28; 408.43 and 250.94; 318.03; 406.12; 635.64; 1.145 mg/mL, for blastula, gastrula, trochophora, veliger and Hippo stage, respectively), adult snails after 24 h of exposure (LC90 of 9.50 and 10.92 mg/mL, respectively) with increased mortality after 7 days of exposure. observation and significant decrease (p<0.05; p<0.01 and p<0.001) in egg mass deposition. At sublethal concentrations, an increase in quantitative and morphological changes in hemocytes was observed, and in the genotoxicity analysis, several degrees of nuclear damage were detected. In addition, ES showed changes in cercariae motility, while FR showed toxicity from a concentration of 1.0 mg/mL. ES showed toxicity to A. salina at the highest concentrations (3.0, 4.0 and 5.0 mg/mL), while the fraction was non-toxic. It is concluded that EP and FR are promising in the fight against schistosomiasis by eliminating the intermediate host and causing alteration and/or mortality to the infectious agent. In addition, ES showed toxicity and FR was non-toxic to A. salina.
